MEL CLASSIFICAÇÃO E QUALIDADE Atividade diastásica (ID) método da C.A.C. (1990), que consiste no uso de uma solução tamponada de amido-iodo-mel mantida em banho-maria (a 40 C), onde se determina o tempo necessário para ser obtido o ponto final específico (correspondente à descoloração da mistura), determinado espectrofotometricamente a 660 nm (absorbância entre 0,240 e 0,200). Quanto mais rápida a descoloração, maior a atividade diastásica do mel, expressa em unidades da escala Gothe por grama de mel, definida como a quantidade de enzima capaz de converter 0,01 g de amido em uma hora a 40ºC

HMF and diastase activity in honeys: A fully validated approach and a chemometric analysis for identification of honey freshness and adulteration Pasias, Ioannis N. ; Kiriakou, Ioannis K. ; Proestos, Charalampos Food Chemistry, 15 August 2017, Vol.229, pp.425-431 A fully validated approach for the determination of diastase activity and hydroxymethylfurfural content in honeys were presented in accordance with the official methods. Methods were performed in real honey sample analysis and due to the vast number of collected data sets reliable conclusions about the correlation between the composition and the quality criteria were exported. The limits of detection and quantification were calculated. Accuracy, precision and uncertainty were estimated for the first time in the kinetic and spectrometric techniques using the certified reference material and the determined values were in good accordance with the certified values. PCA and cluster analysis were performed in order to examine the correlation among the artificial feeding of honeybees with carbohydrate supplements and the chemical composition and properties of the honey. Diastase activity, sucrose content and hydroxymethylfurfural content were easily differentiated and these parameters were used for indication of the adulteration of the honey.
Synephrine A potential biomarker for orange honey authenticity Tette, Patrícia A.S. ; Guidi, Letícia R. ; Bastos, Esther M.A.F. ; Fernandes, Christian ; Gloria, Maria Beatriz A. Food Chemistry, 15 August 2017, Vol.229, pp.527-533 A LC-MS/MS method for synephrine as a biomarker for orange honey authenticity was developed and validated. The sample was extracted with 5% TCA and cleaned up with Florisil providing 83.7% recoveries. Ions transitions for quantification and identification were 168 135.0 and 168 107.0, respectively. The limits of detection and quantification were 0.66 and 1.0 ng/g, respectively. Synephrine was detected in orange honey at levels from 79.2 to 432.2 ng/g, but not in other monofloral honeys. It was also present in some wildflower honeys (9.4 236.5 ng/g), showing contribution of citrus to this polyfloral honey. Results were confirmed by qualitative pollen analysis. No citrus pollen was detected in honey containing synephrine levels 43.8 ng/g, suggesting that synephrine in honey is more sensitive compared to pollen analysis. Synephrine was found in citrus but not in other apiculture flowers. Therefore, synephrine is a botanical marker to differentiate and attest authenticity of orange honey.
