ABSTRACT In the last years, cell cultures in three-dimensional (3D) scaffolds have gained expression in the field of tissue engineering and regenerative medicine as a more physiological and reliable in vitro approach of the in vivo situation. Cell morphology, proliferation, differentiation and regulation were proven to change dramatically from standard two-dimensional (2D) conditions to 3D environment. However, the standard culture in static conditions is not the most suitable for 3D cell culture, due to lack of nutrients and oxygen diffusion towards the scaffolds center. Therefore, dynamic cell culture should be a more physiological approach. This thesis addresses the potential of a multi-compartment container, designed and patented at INEB, to support cell culture in 3D scaffolds under dynamic conditions. The container was conceived to culture cells in 3D fragile hydrogel-like matrices, overcoming the lack of solutions for this type of matrices in the market. As a model system, human mesenchymal stem cells (hmscs) cultured in chitosan (Ch) 3D scaffolds was compared in static and dynamic conditions. The effect of the following parameters was addressed: agitation rate, media feeding regime and the period of cell attachment before dynamic culture. Cell cultures were analyzed for: cell viability, cell proliferation, cell metabolic activity and cell distribution in 3D architecture. In addition, cell capacity to undergo osteogenic differentiation was also analyzed. The data collected clearly suggested higher cell proliferation, more homogenous cell distribution and cell migration towards scaffolds center, under dynamic conditions, when compared to static. Cell metabolic analysis did not reveal major differences between culture conditions. Furthermore, hmscs differentiation into osteogenic lineage appeared to occur earlier when dynamic conditions are used, as suggested by alkaline phosphatase (ALP) activity. Overall, this study highlights some benefits of dynamic culture conditions, particularly using the proposed multi-compartment holder, for 3D culture of hmscs. v
RESUMO Nos últimos anos, a cultura de células em suportes tridimensionais (3D) alcançou uma expressão considerável nos campos da engenharia de tecidos e da medicina regenerativa, apresentando-se como uma abordagem in vitro mais próxima das condições fisiológicas observadas in vivo. Comprovou-se que a morfologia, proliferação, diferenciação e regulação celulares mudam drasticamente quando as células passam da cultura em condições padrão bidimensionais (2D) para um ambiente tridimensional (3D). No entanto, por si só, as condições estáticas não são as mais adequadas para a cultura celular em ambiente 3D, devido à falta de difusão de nutrientes e oxigénio para o centro das matrizes. Por conseguinte, a cultura celular em 3D e em condições dinâmicas constitui uma alternativa mais fisiológica. Esta tese aborda o potencial de um recipiente com múltiplos compartimentos, desenvolvido e patenteado pelo INEB, como suporte para a cultura de células em 3D, sob condições dinâmicas. O recipiente foi concebido para a cultura celular em matrizes 3D frágeis, nomeadamente hidrogéis, no sentido de transpor a falta de soluções no mercado para este tipo de matrizes. Como sistema modelo, a cultura de células estaminais mesenquimatosas humanas (hmscs) em matrizes 3D de quitosano (Ch) foi comparada em condições estáticas e dinâmicas. Ao longo do trabalho, foi abordado o efeito dos seguintes parâmetros: taxa de agitação, regime de mudança de meio e período de adesão das células à matriz, previamente à cultura dinâmica. As culturas foram analisadas, focando em particular a viabilidade celular, proliferação, actividade metabólica e distribuição das células na estrutura 3D. Além disso, foi analisado o potencial de diferenciação osteogénica das hmscs. Os resultados obtidos mostram claramente uma maior proliferação celular, distribuição celular mais homogénea e migração celular para o centro das matrizes de Ch cultivadas sob condições dinâmicas, em comparação com condições estáticas. A análise metabólica não revelou diferenças consideráveis entre os sistemas. Para além disso, a diferenciação das hmscs na linhagem osteogénica ocorreu, aparentemente, mais prematuramente nas amostras cultivadas sob condições dinâmicas, tal como sugerido pela quantificação da atividade da fosfatase alcalina (ALP). De um modo geral, este estudo evidencia os benefícios da cultura celular em condições dinâmicas, particularmente usando o recipiente com múltiplos compartimentos proposto, para cultivar hmscs em 3D. vii
