RESUM Palavras-chave: tetraidro-β-carbolinas, estudo conformacional, atividade tripanossomicida Alcalóides β-carbolínicos e tetraidro-β-carbolínicos naturais e sintéticos têm sido alvo de vários estudos químicos e biológicos. Devido a gama de propriedades farmacológicas, a potencialidade de compostos tetraidro-βcarbolínicos, principalmente como agentes tripanossomicidas e à importância da busca de novos agentes terapêuticos no combate desta doença, realizamos neste trabalho a preparação de uma série de derivados contendo o núcleo tetraidro-βcarbolínico e avaliamos a atividade destes frente às formas epimastigotas de Trypanosoma cruzi e frente à enzima GAPD glicossomal de T. cruzi. Foram realizados também ensaios de toxicidade frente ao microcrustáceo Artemia salina. s derivados tetraidro-β-carbolínicos--carbometóxi--benzossubstituídos foram preparados pela reação de condensação de Pictet-Spengler, catalisada por ácido, do L-triptofano esterificado (6) com uma série de aldeídos. Para cada uma das reações foi obtida a mistura dos isômeros cis (S, S) (a-0a) e trans (R, S) (b-0b). A reação destes compostos, na forma pura ou como mistura de isômeros cis/trans, com as aminas butilamina, isopropilamina, cicloexilamina e pirrolidina forneceu os derivados tetraidro-β-carbolínicos--amido-- benzossubstituídos (-8) e (9a+9b a 56a+56b). Além dos derivados esperados foram obtidos também as β-carbolinas, 5, 58, 59 e 60 resultantes da aromatização do anel piperidínico.
As estruturas dos compostos sintetizados foram confirmadas com base na análise dos dados espectrais de IV, EM e RM e C/DEPT e de técnicas bidimensionais de RM (CSY, MQC e ESY). Constitui-se parte do nosso trabalho ainda, o estudo conformacional dos compostos (a) e (b) através de cálculos computacionais. As estruturas foram otimizadas utilizando a teoria do funcional de densidade (TFD), e o nível de teoria empregado foi o BLYP/6-+G(d). s cálculos foram realizados através do programa Gaussian 98. As análises de B foram feitas utilizando o nível de teoria F/6-G.

ABSTRACT Keywords: tetrahydro-β-carbolinic derivatives, conformational analysis, trypanocidal activity atural and synthetic β-carbolinic and tetrahydro-β-carbolinic alkaloids have been object of several chemical and biological studies due to their large spectrum of pharmacological proprieties. The importance of this class of compounds and, specifically its tripanocidal activity, led us to synthesize a series of tetrahydro-βcarbolinic derivatives and to evaluate their activity against the epimastigote forms of Trypanosona cruzi and against the enzyme GAPD of T. cruzi. Additionally, the toxicity against Artemia salina was tested. The tetrahydro-β-carbolinic -carbomethoxy--benzosubstituted derivatives were prepared by the condensation of Pictet-Splenger, under acid catalysis, of the L-tryptophan methyl ester (6) with a series of aldehydes to afford a mixture of isomers cis (R, S) (a-0a) and trans (R, S) (b-0b). The reaction of these compounds, in the pure form or as mixture of isomers, with the amines butilamine, i-propylamine, cyclohexylamine and pyrrolidine produced the tetrahydro-β-carbolinic -amide--benzosubstituted derivatives (-8) and (9a+9b to 56a+56b). Some unexpected products from aromatiztion (, 5, 58, 59 and 60) were also observed. The synthesized compounds structure were confirmed on the basis of their spectral data (IR, MS, and C MR, CSY and MQC).
In order to explain some aspects related to the MR data from cis and trans isomers conformational studies by computational methods were carried out. The structures were optimized using density functional theory. The BLYP hybrid functional was used in this investigation. The 6-+G(d) basis set was used in these calculations. The natural bond orbital (B) calculation was performed using F/6-G level of theory.
