5 Resumo O citosesqueleto de actina é um constituinte essencial das células eucariotas, produzindo forças de tensão que moldam a membrana das células e constituindo o suporte estrutural necessário à ancoragem e movimento de organelos e vesículas. Um grande conjunto de proteínas de ligação à actina (ABPs) controla, de uma forma elaborada, a dinâmica dos filamentos deste citoesqueleto, regulando processos como o tráfego polarizado de vesículas, a migração celular e a citocinese. Por sua vez, num contexto multicelular, interacções célula-célula e célula-matriz, modulam o estado e actividade de muitos dos constituintes básicos do citosqueleto de actina. Em particular, os processos que regulam a dinâmica dos filamentos de actina parecem responder a vias de sinalização que regulam o crescimento, proliferação e morte celulares, muitas das quais se encontram desreguladas em vários tipos de cancro. Apesar das evidentes alterações estruturais sofridas pelo citoesqueleto de actina em células tumorais, pouco é de facto conhecido acerca de como é que estas alterações são capazes de contribuir para distorções do crescimento de tecidos e órgãos no contexto de um organismo. Estudos recentes no organismo modelo Drosophila melanogaster ( mosca-dafruta ) têm contribuído para a compreensão de muitos processos morfogenéticos e de padronização de tecidos epiteliais, debruçando-se, em particular, sobre o estudo dos discos imaginais. Os discos imaginais são pequenos tecidos de origem ectodérmica que se encontram interiorizados na larva de Drosophila, sofrendo processos de morfogénese, crescimento e diferenciação complexos, que após a metamorfose, darão origem aos membros articulados do insecto adulto. Em particular, o disco imaginal da asa constitui um modelo epitelial, sendo constituído por duas camadas de células, uma colunar e outra cuboidal. Dada a fácil manipulação desta estrutura epitelial e devido ao enorme conhecimento adquirido sobre a sua padronização e crescimento, os disco imaginal da asa foi escolhido neste estudo como um modelo de organização epitelial. Estudos sobre o papel do heterodímero Capping Protein (CP) durante a morfogénese do disco imaginal da asa revelaram, sem precedentes, dados que apontam o citoesqueleto de actina como um importante supressor do crescimento no v

6 contexto de um tecido epitelial. O heterodímero CP é constituindo por duas subunidades - (Cpa) e (Cpb) - sendo capaz de bloquear a polimerização de actina, impedindo a adição e perda de monómeros ao extremo barbado (+) dos filamentos. Mutações nos genes codificantes das subunidades (cpa) ou (cpb) de CP, ambas gerando um heterodímero não funcional, dão origem a diferentes respostas celulares consoante o tecido epitelial no qual se desenvolvem. Por exemplo, no disco imaginal da asa, grupos de células mutantes (clones) para cpa ou cpb sofrem extrusão e apoptose na porção mais distal, que dará origem à lâmina alar, não produzindo qualquer perda de integridade epitelial nas regiões mais proximais do disco. No entanto, o silenciamento génico de cpa ou cpb (CP), numa região abrangente de toda a região distal do disco imaginal da asa, resulta no desenvolvimento neoplástico do tecido. Curiosamente, à semelhança do que sucede com mutações em genes associados à regulação de processos de endocitose, incluindo o da sintaxina avalanche (avl) e rab5, o silenciamento génico de cpb por RNA de interferência em células epiteliais de Drosophila, induz uma acumulação do determinante de polaridade apical Crumbs (Crb) associada ao desenvolvimento de tumores neoplásticos. Por outro lado, o produto de um inserto transgénico de cpa marcado por um epítopo de hemaglutinina (HA), sendo capaz de suprimir o fenótipo de mutantes cpa, encontra-se localizado em pontuações de tipo vesicular. Estes resultados são preliminares aos resultados apresentados nesta tese, sugerindo que CP possa estar directamente envolvido no movimento ou formação de vesículas de endocitose, ou que a rede de filamentos de actina originada sobre a acção do heterodímero CP possa ser responsável por esses processos. O objectivo principal deste projecto de investigação consistiu em esclarecer o envolvimento directo ou indirecto de CP no tráfego vesicular de endocitose bem como em averiguar se a acumulação do determinante de polaride Crb, associada a sobreproliferação e formação de tumores neoplásticos em células mutantes CP, é uma consequência de defeitos no tráfego vesicular de endocitose. Os resultados apresentados nesta tese revelaram uma associação estreita entre CP e endossomas primários, marcados pela proteína Rab5, sugerindo que CP possa contribuir para a regulação da dinâmica de formação e fusão deste tipo de vi

7 endossomas. Tal como esperado, em células que sofrem um decaimento da expressão de cpb por RNAi, foi observada uma acumulação de Crb em estruturas de tipo vesicular. Para além da acumulação de Crb, este estudo revela ainda que o silenciamento génico de cpb resulta numa expressiva acumulação de outras proteínas membranares, nomeadamente do receptor de sinalização Notch e da proto-caderina Fat. Estes resultados sugerem que a função de CP é relevante para prevenir um defeito generalizado na internalização de receptores membranares por endocitose, o qual poderá causar perturbações em várias vias de sinalização mediadas por esses receptores. Por exemplo, à semelhança do que ocorre em tecido mutante para o gene avl, a perda de função de CP parece induzir uma interrupção do tráfego vesicular de Notch, impedido o seu processamento proteolítico em endossomas secundários. De facto, este estudo demonstra a ocorrência de uma disfunção da via de sinalização Notch em células desprovidas de CP por RNAi, uma vez que estas apresentam uma redução da expressão de genes alvos da via Notch no disco imaginal da asa, como cut e m8(espl). Neste estudo foi ainda verificado que, mediante o silenciamento génico de cpb por RNAi, a acumulação de Fat é concomitante com o desenvolvimento de defeitos de polaridade planar, tais como a desorientação dos pelos da asa, bem como com o crescimento anormal de uma região particular do disco imaginal, sugerindo uma perturbação da actividade supressora de tumores desta proto-caderina aquando da disfunção do heterodímero CP. Este estudo revela ainda que a sobrexpressão do domínio citoplasmático de Crb no disco imaginal da asa induz perda de polaridade celular e o crescimento anormal de uma região particular, destinada a formar as estruturas de articulação da asa com o tórax. Por outro lado, clones de células epiteliais sobrexpressando o domínio citoplasmático de crb sofrem apoptose, sendo esta induzida pelas células normais circum-adjacentes de acordo com um processo de competição celular. No entanto, tal como sucede com clones de células mutantes para as subunidades de CP, os clones que sobrexpressam o domínio citoplasmático de crb sofrem uma extrusão preferencial na região mais distal do disco imaginal da asa, a qual dará origem à lâmina alar. Estes resultados sugerem que, ao longo de um tecido epitelial as células não respondem da mesma maneira aos níveis deste determinante de polaridade e vii

8 que os níveis de Crb são necessários para a regulação da proliferação celular em domínios restritos do disco imaginal da asa. Inesperadamente, este estudo revelou ainda uma interacção genética entre CP e o determinante de polaridade apkc, na regulação do tamanho e forma de células epiteliais. Os resultados apresentados nesta tese sugerem que CP e apkc possam contribuir paralelamente para a regulação da dinâmica do citoesqueleto de actina, sendo cruciais para a estruturação da forma colunar de células epiteliais. No seu conjunto, os dados apresentados nesta tese apontam para o impacto crucial do citoesqueleto de actina na regulação do crescimento e forma celular no contexto de um tecido epitelial. 9 Abstract The actin cytoskeleton is an essential constituent of eukaryotic cells. In addition to controlling numerous processes, such as the generation and maintenance of cell morphology and polarity, contractility and cell division, the actin microfilament system provides the structural support necessary for defining the precise location and movement of organelles and vesicles. A complex array of actin binding proteins (ABPS) tightly controls the dynamics of actin filament assembly, disassembly and organization within the cell. Recent studies in Drosophila have provided unprecedented evidence suggesting that capping of specific actin filament pools is critical to restrain epithelial growth. The Capping Protein subunits a and (Cpa and Cpb) form a conserved heterodimer (CP), which restricts actin polymerization by preventing the addition and loss of monomers to the filaments barbed (+) end. Interestingly, similar to mutations in genes associated with the endocytic pathway, including the syntaxin avalanche (avl) and rab5, loss of capping protein (cpa), in Drosophila, results in Crumbs (Crb) accumulation coupled with overproliferation and development of neoplastic tumours. Additionally, an HA-Tagged form of cpa, which can fully rescue the loss of cpa, localizes to vesicle-like puncta, suggesting that CP is directly involved in vesicle movement. The main goal of this research project was to determine whether CP is involved in vesicle trafficking, either directly or indirectly and to analyse whether Crumbs (Crb) accumulation, due to impaired vesicle trafficking, is the cause of overproliferation and neoplastic transformation of CP mutant tissue. The results presented in this thesis indicate that CP promotes endocytosis of membrane-linked proteins: I found that besides accumulating Crb, tissue depleted of CP by RNAi accumulates other surface receptors, such as Notch and Fat. These defects are unlikely to be due to transcriptional upregulation of these molecules since Notch is unable to signal in these cells, a feature that has also been observed in avl mutant tissue. Moreover, my results show that CP co-localizes with the early endosome marker Rab5. Furthermore, overexpressing the two cytoplasmic actin genes also results in Crumbs accumulation and hinge overgrowth. This suggests that the actin ix

10 capping activity of CP directly regulates trafficking of early endosomes to promote receptor clearance from the plasma membrane. Similar to tissue depleted of CP, I found that wing epithelia overexpressing crb overgrow predominantly in the presumptive distal hinge, being concomitant with upregulation of wingless expression. Furthermore, clones of crb overexpressing cells look to be out-competed by surrounding tissue through a process of cell competition, and, as in CP mutant cells, cell extrusion occurs preferentially in the distal wing epithelium, where the wing blade will form. This suggests that cells do not respond identically to Crb levels along the proximal-distal axis, and that Crb levels are critical to restrain cell growth. Although overexpression of a kinase-dead form of apkc, which can reduce the activity of overexpressed crb, could not suppress the CP depletion phenotype, accumulation of membrane-linked proteins, including Crb, could at least be partially responsible for the overgrowth of CP depleted tissue. Unexpectedly, this work has revealed a genetic interaction between CP and the polarity determinant apkc in the maintenance of epithelium cell polarity and the regulation of cell size and shape. My results suggests that CP is crucial to promote the formation of an actin network that provides tension and prevents cell extrusion when AJ stability is compromised, such as in the case of apkc loss of function. Moreover, CP and apkc might cooperatively regulate actin dynamics to promote the columnar cell shape of epithelial cells. In the near future, understanding how the actin cytoskeleton is regulated to promote vesicle trafficking will help us understand how this cellular constituent impinges on growth to maintain tissue homeostasis and prevent tumoral behaviour.
