2 SureStep ENGLISH hg ombo (II) Pregnancy est For In Vitro Diagnostic Use INENDED USE For the rapid detection of human chorionic gonadotropin (hg) in urine and serum specimens. his test kit is used to obtain a visual, qualitative result and is intended for professional and laboratory use only. SUMMARY Human chorionic gonadotropin (hg) is a glycoprotein hormone secreted by the developing placenta shortly after fertilization. In normal pregnancy, hg can be detected in serum as early as 7 days following conception (ref.1-4). he concentration of hg continues to rise rapidly, frequently exceeding 100 miu/ml by the first missed period (ref. 1-5) and peaking in the IU/ml range by weeks into pregnancy. he appearance of hg soon after conception and its subsequent rise in concentration during early gestational growth make it an excellent marker for the early detection of pregnancy. he SureStep hg ombo (II) Pregnancy est is a rapid test that detects the presence of hg in a qualitative format sensitive to 20 miu/ml in urine and 10 mlu/ml in serum. he test utilizes monoclonal antibodies to selectively detect elevated levels of hg in serum and urine specimens. he immunological specificity of the test kit virtually eliminates cross-reactivity interference from the structurally related glycoprotein hormones hfsh, hlh and hsh at physiological levels. PRINIPLE he SureStep hg ombo (II) Pregnancy est is a chromatographic immunoassay (IA) for the rapid qualitative determination of hg in urine and serum. he membrane is precoated with anti-alpha hg capture antibody on the test line region and goat anti-mouse on the control line region. During testing, the urine or serum specimen is allowed to react with colloidal gold particles coated with anti-beta hg monoclonal antibody. he mixture then moves along the membrane chromatographically by capillary action. For a positive result, a pink-colored line with a specific antibody-hg-antibody-colloidal gold particle complex will form on the membrane in the test line region. Absence of a pink-colored line in the test line region indicates a negative result. he appearance of a colored line in the control region serves as verification that sufficient volume has been added and that proper flow was obtained. Specimens collected at any time of day may be used; however, the first morning urine generally contains the highest concentration of hormone. Urine specimens may be refrigerated at 2-8 (36-46 F) and stored for up to 72 hours prior to testing. If samples are refrigerated, they must be equilibrated to room temperature before testing. Urine samples exhibiting visible precipitates should be filtered, centrifuged or allowed to settle, prior to testing. Serum Samples: No special preparation of the patient serum is required. Serum not assayed immediately must be stored refrigerated 2-8 (36-46 F) for up to 72 hours or frozen -20 (-4 F) for up to 3 months. Do not freeze and thaw sample repeatedly. Grossly hemolyzed samples should not be used. ES PROEDURE Review Specimen ollection instructions. If frozen or refrigerated, test devices, patient samples, and controls should be brought to room temperature (68-86 F) prior to testing. Do not open pouches until ready to perform the assay. 1. Remove the test device from its protective pouch (devices must be brought to room temperature before opening the pouch to avoid moisture condensation on the membrane). Label the device with patient or control identifications. Place device on a clean flat surface. 2. Draw sample to the line marked on the pipette (approximately 0.2 ml). Dispense the entire contents into the sample well. For each sample or control, use a separate pipette and device. 3. Wait for pink-colored lines to appear. Depending on the concentration of hg, positive results may be observed as soon as 40 seconds. o confirm a negative result, a reaction time of 5 minutes is required. When testing a urine specimen, the first morning specimen generally contains the highest concentration of hg. LIMIAIONS OF HE PROEDURE A number of conditions other than pregnancy including trophoblastic disease and certain nontrophoblastic neoplasms can cause elevated levels of hg. he test is not intended to diagnose conditions other than pregnancy. If a urine specimen is too dilute it may not contain representative levels of hg. If pregnancy is still suspected, a first morning urine should be obtained from the patient hours later and tested. As with all diagnostic tests, a definitive clinical diagnosis should not be based on the result of a single test, but should only be made by a physician after all clinical and laboratory findings have been evaluated. 5 Immunologically interfering substances such as those used in antibody therapy treatments may invalidate the test result. Some samples containing very high levels of hg (~ 200,000 mlu/ml) may yield a test line with a color intensity lighter than expected. When high dose hook effect is suspected, a 1:10 dilution of the specimen with distilled water is recommended. he test should then be repeated with the diluted specimen. he presence of heterophile antibodies or non-specific protein binding may cause false-positive results in sensitive immunoassays. If a qualitative interpretation is inconsistent with the clinical evidence, results should be confirmed by an alternative hg method. EXPEED VALUES Healthy men and healthy non-pregnant women do not have hg levels detectable by the SureStep hg ombo (II) Pregnancy est. For pregnant women, hg levels of 100 miu/ml can be reached on the day of the first missed menstrual period. hg levels peak about 8-10 weeks after the last menstrual period and then decline to lower values for the remainder of the pregnancy. Following delivery, hg levels rapidly decrease and usually return to normal within days after parturition. QUALIY ONROL A procedural control is included in the test. A colored line appearing on the control region () is considered an internal positive procedural control, indicating proper performance. A clear background in the result window is considered an internal negative procedural control. If the test is working properly and has been performed correctly, the background will clear giving a distinct result. It is recommended that two levels of control specimens be used with each new kit, however, each laboratory should follow their state and local requirements. 7 Interference esting he following substances were added to hg-free urine (negative sample) and 20 miu/ml hg-spiked urine (positive sample) and tested in the assay. No interference was observed with the following substances at the concentrations listed below. In addition, the effect of urine ph from 5 to 9 was tested at these hg concentrations. he ph of the urine did not affect the outcome of the result. 9 SureStep DEUSH hg ombo (II)- Schwangerschaftstest Zur Anwendung in der In-vitro-Diagnostik VERWENDUNGSZWEK Zum Schnellnachweis von humanem horiongonadotropin (hg) in Urin- und Serumproben. Dieses estkit dient der Erhebung eines visuellen qualitativen Ergebnisses und ist ausschließlich für die Anwendung durch medizinisch geschultes Personal im Rahmen des Klinik- oder Ambulanzbetriebs bestimmt. ZUSAMMENFASSUNG Das humane horiongonadotropin (hg) ist ein bereits kurz nach der Befruchtung der Eizelle von der Plazenta sezerniertes Glykoproteinhormon. Bei normalem Schwangerschaftsverlauf ist hg bereits 7 age nach der Konzeption nachweisbar (Lit. 1-4). In der Folge steigt die hg-konzentration weiter rasch an und liegt zu dem Zeitpunkt, an dem die Menstruation erstmals ausbleibt, vielfach bereits über 100 miu/ml (Lit. 1-5). Maximale hg-konzentrationen von IU/ml werden zwischen der 10. und 12. Schwangerschaftswoche erreicht. Da hg bereits kurz nach der Konzeption nachweisbar ist und im weiteren Verlauf der Frühschwangerschaft kontinuierlich ansteigt, ist dieses Hormon ein hervorragender Marker für die Früherkennung einer Schwangerschaft. Der SureStep hg ombo (II)-Schwangerschaftstest ist ein Schnelltest, der hg- Konzentrationen ab 20 miu/ml im Urin und 10 mlu/ml im Serum qualitativ nachzuweisen vermag. Der selektive Nachweis erhöhter hg-konzentrationen in Urin und Serum basiert dabei auf dem Einsatz monoklonaler Antikörper. Dank der immunologischen Spezifität des ests sind Interferenzen infolge Kreuzreaktivität mit strukturell verwandten Glykoproteinhormonen - dem humanen follikelstimulierenden Hormon (hfsh), dem humanen luteinisierenden Hormon (hlh) und dem humanen hyreotropin (hsh) - in physiologischen Konzentrationen praktisch ausgeschlossen. ESPRINZIP Der SureStep hg ombo (II)-Schwangerschaftstest ist ein chromatografischer Immunoassay (IA) zum qualitativen Schnellnachweis von hg in Urin und Serum. Die Membran ist im Bereich des estfeldes mit Anti-alpha-hG-apture-Antikörpern und im Bereich des Kontrollfeldes mit Anti-Maus-Antikörpern (Ziege) beschichtet. Während des estverlaufs reagiert die aufgebrachte Urin- bzw. Serumprobe mit kolloidalen, mit monoklonalen Anti-beta-hG-Antikörpern beschichteten Goldpartikeln. Diese Mischung wandert durch Kapillaraktivität chromatografisch entlang der Membran. Ist der est positiv, kommt es auf der Membran im estfeld durch spezifische Komplexbildung zwischen Anti-hG-Antikörpern und kolloidalen Goldpartikeln zur Entstehung einer rosafarbenen Linie. Bildet sich keine DE 1

10 rosafarbene Linie im estfeld, ist das Ergebnis negativ. Die Entstehung einer Farblinie im Kontroll- und im Referenzfeld zeigt an, dass das Probenvolumen ausreichend war und der Kapillarfluss wie vorgesehen erfolgte. AUFBEWAHRUNG UND SABILIÄ Bei gekühlter Aufbewahrung oder Aufbewahrung bei Raumtemperatur (zwischen 2 und 30 ) in der ungeöffneten Originalverpackung ist der est bis zum angegebenen Verfalldatum stabil. Die Probennahme kann zu jeder beliebigen ageszeit erfolgen; allerdings sind im ersten Morgenurin die höchsten Hormonkonzentrationen nachweisbar. Vor Durchführung des ests können Urinproben gekühlt (2 bis 8 ) bis zu 72 Stunden gelagert werden. Gekühlte Proben sind vor Verwendung auf Raumtemperatur zu equilibrieren. In der Urinprobe eventuell sichtbare Präzipitate sollten vor der Verwendung herausgefiltert, abzentrifugiert oder durch Sedimentation entfernt werden. Serumproben: Serumproben bedürfen keiner besonderen Vorbereitung. Serum, das nicht sofort getestet wird, ist gekühlt (2 bis 8 ) bis zu 72 Stunden und gefroren (-20 ) bis zu 3 Monate haltbar. Proben nicht wiederholt einfrieren und auftauen. Stark hämolytische Proben sind zu verwerfen. ESVERFAHREN Die unter Probennahme angeführten Hinweise beachten. Gekühlte oder tiefgekühlte esteinheiten, Patientenproben und Kontrollen sind vor Durchführung des ests auf Raumtemperatur (20 bis 30 ) zu equilibrieren. Die Folienverpackung erst unmittelbar vor Durchführung des ests öffnen. 1. Die esteinheit aus der Schutzverpackung nehmen. (Um eine kondensationsbedingte Feuchtigkeitsbildung auf der Membran zu verhindern, die Verpackung erst öffnen, wenn der est Raumtemperatur erreicht hat.) Den est jeweils mit einem Patienten- bzw. Kontrolletikett kennzeichnen. Den est auf eine saubere, flache Unterlage legen. 2. Probenflüssigkeit bis zu der auf der Pipette angegebenen Markierung aufziehen (ca. 0,2 ml). Den gesamten Pipetteninhalt in die Probenvertiefung geben. Für jede Probe bzw. Kontrolle eine neue Pipette und einen neuen est verwenden. DE 2

11 3. Warten, bis rosafarbene Linien sichtbar werden. Je nach hg-konzentration sind positive estergebnisse bereits nach 40 Sekunden ablesbar. Ein negatives estergebnis gilt erst nach einer Reaktionszeit von 5 Minuten als gesichert. Nach 10 Minuten kann das estergebnis nicht mehr sicher ausgewertet werden. Die Farbintensität der estlinie () kann mit der in der Probe vorhandenen hg-konzentration variieren. Allerdings können mittels qualitativer ests weder ein quantitativer Wert noch das Ausmaß des hg-anstiegs erhoben werden. NEGAIV: Es erscheint nur eine rosafarbene Linie im Kontrollfeld (). Im estfeld () bildet sich keine Farblinie aus. UNGÜLIG: Das Fehlen einer rosafarbenen Linie im Kontrollfeld ist als Hinweis auf einen Verfahrensfehler oder auf unbrauchbar gewordene estreagenzien zu werten. Den est mit einer neuen esteinheit wiederholen und prüfen, ob im Kontrollfeld eine rosafarbene Linie entsteht. Positiv Negativ Ungültig HINWEISE ZUR AUSWERUNG DER ESERGEBNISSE Liegt ein negatives estergebnis vor, obwohl eine Schwangerschaft vermutet wird, sollte der est mit einer 48 bis 72 Stunden später entnommenen Probe wiederholt oder ein quantitativer est durchgeführt werden. Bei Verwendung von Urinproben ist zu beachten, dass der erste Morgenurin üblicherweise die höchsten hg- Konzentrationen enthält. GRENZEN DES VERFAHRENS Neben einer Schwangerschaft können auch etwa trophoblastäre Erkrankungen sowie bestimmte nicht-trophoblastäre Neoplasmen Ursache für erhöhte hg- Konzentrationen sein. Dieser est ist ausschließlich für die Diagnose einer Schwangerschaft, nicht aber für die Diagnose von Erkrankungen bestimmt. DE 3

12 Bei zu stark verdünnten Urinproben liegen die hg-konzentrationen möglicherweise unter der Nachweisgrenze des ests. Wird dennoch eine Schwangerschaft vermutet, sollte Stunden später eine Probe aus dem ersten Morgenurin entnommen und getestet werden. Wie bei allen diagnostischen ests sollte die endgültige klinische Diagnose nicht von einem Einzelergebnis abgeleitet, sondern ausschließlich von einem Arzt nach Auswertung sämtlicher klinischer Untersuchungsergebnisse und Laborbefunde gestellt werden. Immunologische Störsubstanzen wie etwa Substanzen, die in einer Antikörpertherapie zur Anwendung kommen, können die estergebnisse beeinträchtigen. Einige Proben mit sehr hohen hg-konzentrationen (~ mlu/ml) können zur Entstehung einer heller als erwarteten Farblinie führen. Wird ein High-Dose-Hook- Effekt vermutet, sollte die Probe im Verhältnis 1:10 mit destilliertem Wasser verdünnt und der est mit der verdünnten Probe wiederholt werden. Bei hochempfindlichen Immunoassays kann das Vorliegen heterophiler Antikörper bzw. eine unspezifische Proteinbindung zu falsch-positiven Ergebnissen führen. Entspricht die qualitative estauswertung nicht dem klinischen Gesamtbild, sind die Ergebnisse unter Verwendung einer alternativen Methode zur hg-bestimmung zu bestätigen. ERWAREE WERE Bei gesunden Männern und gesunden nicht schwangeren Frauen sind mit Hilfe des SureStep hg ombo (II)-Schwangerschaftstests keine hg-konzentrationen nachweisbar. Bei schwangeren Frauen können bereits am ersten ag, an dem die Menstruation ausbleibt, hg-konzentrationen von 100 miu/ml erreicht werden. Maximale hg-konzentrationen werden 8 bis 10 Wochen post menstruationem erreicht, im weiteren Verlauf der Schwangerschaft fällt die hg-konzentration wieder auf niedrigere Werte ab. Innerhalb weniger age nach der Entbindung sinkt die hg-konzentration gewöhnlich wieder auf den Normalwert. QUALIÄSKONROLLE Der est verfügt über eine testimmanente Verfahrenskontrolle. Eine Farblinie im Kontrollfeld () gilt als positive interne Verfahrenskontrolle und als Hinweis auf einen ordnungsgemäßen establauf. Ein unverfärbter Hintergrund im Ergebnisfenster gilt als negative interne Verfahrenskontrolle. Bei ordnungsgemäßem estverlauf und sachgemäßer Durchführung ist der Hintergrund unverfärbt und sorgt so für ein eindeutiges Ergebnis. Es empfiehlt sich, mit jedem neuen Kit Kontrollproben in zwei Konzentrationen mitzuführen, generell ist hierbei jedoch den jeweils geltenden örtlichen Bestimmungen Folge zu leisten. Als Kontrollmaterialien stehen etwa die SureStep hg -Positiv- und Negativkontrollen zur Verfügung (Best.-Nr. 6009). LEISUNGSMERKMALE Sensitivität Der SureStep hg ombo (II)-Schwangerschaftstest weist hg- Urinkonzentrationen ab 20 miu/ml und hg-serumkonzentrationen ab 10 mlu/ml (kalibriert gemäß 3. IRP) nach und zeigt ab diesen Konzentrationen eine rosafarbene Linie im estfeld () an. Auch hg-urinkonzentrationen unter 20 miu/ml und hg-serumkonzentrationen unter 10 mlu/ml können zu schwach positiven Ergebnissen führen. Zur Beurteilung der Sensitivität des SureStep hg ombo (II)-Schwangerschaftstests bei niedrigen hg-konzentrationen wurden folgende Untersuchungen durchgeführt: DE 4

13 A. Urinproben: Urinproben von fünf nachweislich nicht schwangeren Frauen wurden auf hg-konzentrationen von 0, 5 und 20 miu/ml aufgestockt. Insgesamt wurden 120 Proben verblindet und mit dem SureStep hg ombo (II)- Schwangerschaftstest getestet. Die Ergebnisse sind in abelle 1 zusammengefasst. Bei Konzentrationen von 300 mlu/ml für hlh, 1000 miu/ml für hfsh und 1000 µiu/ml für hsh waren alle estergebnisse negativ. Genauigkeit: Ergebnisse von Korrelationsstudien A. Urinproben: Einhundert klinische Urinproben wurden sowohl mit dem SureStep hg ombo (II)-Schwangerschaftstest als auch mit einem handelsüblichen hg-est mit Ein-Schritt-echnologie analysiert. 14 Interferenztests hg-freier Urin (negative Probe) und eine auf eine hg-konzentration von 20 miu/ml aufgestockte Probe (positive Probe) wurden mit den im Folgenden angeführten Substanzen versetzt und getestet. Dabei kam es in den jeweils angegebenen Konzentrationen zu keinen Interferenzen. Darüber hinaus wurden die Auswirkungen eines Urin-pH zwischen 5 und 9 bei oben angeführten hg-konzentrationen getestet. Der ph des Urin hatte keine Auswirkungen auf die Ergebnisse. 16 SureStep ESPAÑOL Prueba del embarazo hg ombo (II) Para uso diagnóstico in vitro USO PREVISO Para la detección rápida de gonadotropina coriónica humana (hg) en muestras de orina y suero. Esta prueba se usa para obtener un resultado visual cualitativo y está destinada exclusivamente para uso profesional y en laboratorio. RESUMEN La gonadotropina coriónica humana (hg) es una hormona glucoproteica secretada por la placenta en desarrollo poco después de la fertilización. En el embarazo normal, se puede detectar hg en suero ya a los 7 días de la concepción (1-4). La concentración de hg continúa aumentando rápidamente, superando con frecuencia las 100 mui/ml en el momento de la primera falta (1-5) y alcanzando su máximo en torno a las UI/ml a las semanas de embarazo. La aparición de hg poco tiempo después de la concepción y el posterior aumento de su concentración al principio del crecimiento gestacional convierten a esta hormona en un excelente marcador para la detección precoz del embarazo. La prueba de embarazo SureStep hg ombo (II) es una prueba rápida que detecta cualitativamente la presencia de hg con una sensibilidad hasta 20 mui/ml en orina y 10 mui/ml en suero. La prueba utiliza anticuerpos monoclonales para detectar selectivamente las concentraciones elevadas de hg en las muestras de suero y orina. La especificidad inmunológica de la prueba elimina prácticamente la interferencia por reacción cruzada de las hormonas glucoproteicas estructuralmente relacionadas, hfsh, hlh y hsh, en concentraciones fisiológicas. PRINIPIO DE LA PRUEBA La prueba de embarazo SureStep hg ombo (II) es un inmunoensayo cromatográfico (IA) para la determinación rápida cualitativa de hg en orina y suero. La membrana está recubierta previamente con anticuerpo de captura antialfa-hg en la zona de la línea de prueba y de cabra antirratón en la zona de la línea de control. Durante la prueba, se permite que la muestra de orina o suero reaccione con las partículas de oro coloidal recubiertas con anticuerpo monoclonal anti-betahg. La mezcla se desplaza entonces a lo largo de la membrana por cromatografía mediante acción capilar. Para que el resultado sea positivo, se formará sobre la membrana una línea de color rosa con un complejo específico anticuerpo-hganticuerpo-partícula de oro coloidal en la zona de la línea de prueba. La ausencia de la línea de color rosa en la zona de la línea de prueba indica un resultado negativo. Se pueden usar las muestras obtenidas a cualquier hora del día; sin embargo, la primera orina de la mañana contiene normalmente la concentración más alta de la hormona. Las muestras de orina se pueden refrigerar a 2-8 (36-46 F) y almacenarse hasta un máximo 72 horas antes de la prueba. Si las muestras están refrigeradas, se dejará que alcancen la temperatura ambiente antes de la prueba. Extraiga la muestra con la pipeta hasta la línea marcada (aproximadamente 0,2 ml). Ponga todo el contenido en el pocillo de la muestra. 18 3. Espere a que aparezcan las líneas de color rosa. Dependiendo de la concentración de hg, los resultados positivos pueden verse ya a los 40 segundos. Para confirmar un resultado negativo, es necesario dejar transcurrir un tiempo de reacción de 5 minutos. No deben interpretarse los resultados una vez transcurridos más de 10 minutos. 19 LIMIAIONES DEL PROEDIMIENO Hay varias situaciones distintas del embarazo, como la enfermedad trofoblástica y algunas neoplasias no trofoblásticas, que pueden elevar las concentraciones de hg. La prueba no está diseñada para diagnosticar situaciones distintas del embarazo. Si la muestra de orina está demasiado diluida, es posible que no contenga niveles representativos de hg. Si, aún así, se sospecha el embarazo, se deberá obtener la primera orina de la mañana de la paciente horas más tarde y repetir la prueba. omo sucede con todas las pruebas diagnósticas, el médico no debe establecer el diagnóstico clínico definitivo sobre la base de los resultados de una prueba aislada, sino después de evaluar todos los resultados clínicos y analíticos. Las sustancias que provocan interferencias inmunológicas, como las que se utilizan en las terapias con anticuerpos, pueden invalidar los resultados de la prueba. Las muestras que contienen concentraciones muy altas de hg (~ mui/ml) pueden dar una línea de prueba con un color más claro de lo esperado. uando se sospeche un efecto de gancho por dosis alta, se recomienda diluir la muestra 1:10 con agua destilada. La prueba se debe repetir con la muestra diluida. La presencia de anticuerpos heterófilos o unión inespecífica de proteínas puede provocar resultados falsos positivos en los inmunoensayos sensibles. Si la interpretación cualitativa no es compatible con los datos clínicos, se deben confirmar los resultados con un método de hg alternativo. EXPEAIVA DE RESULADOS Los varones sanos y las mujeres no embarazadas no tienen concentraciones detectables de hg con la prueba del embarazo SureStep hg ombo (II). En las mujeres embarazadas, se pueden alcanzar concentraciones de hg de 100 mui/ml en el día de la primera falta. La hg alcanza su máximo a las 8-10 semanas después de la primera falta y después decae a valores inferiores durante el resto del embarazo. Después del parto, las concentraciones de hg disminuyen rápidamente y, en general, vuelven a la normalidad pocos días después del parto. ONROL DE ALIDAD En la prueba se incluye un procedimiento de control. La línea de color que aparece en la zona del control () se considera un control interno positivo del procedimiento e indica un comportamiento adecuado. Un fondo transparente en la ventana de resultados se considera un control interno negativo del procedimiento. Si el funcionamiento de la prueba es el adecuado y se ha realizado correctamente, el fondo se borrará, dando un resultado nítido. Se recomienda usar dos niveles de muestras de control con cada nuevo equipo, aunque cada laboratorio debe seguir los requisitos estatales y locales correspondientes. Para ello, recomendamos usar los controles positivo y negativo de hg SureStep (nº de catálogo 6009). ARAERÍSIAS DE FUNIONAMIENO Sensibilidad La prueba del embarazo SureStep hg ombo (II) detecta concentraciones de hg en orina iguales o mayores de 20 mui/ml y concentraciones séricas iguales o mayores de 10 mui/ml (calibrado de acuerdo con la 3ª IRP) indicadas por la aprarición de una línea de color rosa en la zona de la prueba (). Además, las muestras de orina que contienen menos de 20 mui/ml de hg y las muestras de suero que contienen menos de 10 mui/ml de hg también pueden producir un resultado positivo débil. Para evaluar la sensibilidad de la prueba del embarazo SureStep con concentraciones más bajas de hg, se realizaron los siguientes experimentos: ES 4

RÉSUMÉ La gonadotrophine chorionique humaine (hg) est une hormone glycoprotéique produite par le placenta en cours de développement tout de suite après la fécondation. Dans les grossesses normales, la hg peut être détectée dans le sérum dès le 7ème jour qui suit la fécondation (réf. 1-4). La concentration de hg continue d augmenter rapidement, dépassant souvent 100 mui/ml lors de l absence des premières règles (réf. 1-5) et atteignant UI/ml à partir des èmes semaines de grossesse. PRINIPE Le test de Grossesse SureStep hg ombo (II) est un immuno-essai chromatographique (IA) pour la détermination qualitative rapide de la hg dans l urine et le sérum. La membrane est préalablement enduite d anticorps anti-alpha de détection de la hg dans la partie de la ligne de test, ainsi que de chèvre antisouris dans la partie de la ligne de contrôle. Pendant le test, l échantillon d urine ou de sérum réagit avec les particules d or colloïdal revêtues d anticorps monoclonal hg anti-bêta. Le mélange se déplace ensuite de manière chromatographique le long de la membrane par action capillaire. En cas de résultat positif, une ligne rose, avec un ensemble spécifique anticorps-hg-anticorps-particules d or colloïdal se formera sur la membrane dans la partie de la ligne de test. Il est possible d utiliser des échantillons prélevés à n importe quel moment de la journée ; toutefois, la première urine du matin contient généralement les concentrations les plus élevées de l hormone. Les échantillons d urine peuvent être réfrigérés à 2-8 (36-46 F) et stockés durant un maximum de 72 heures avant le test. Si les échantillons ont été réfrigérés, ils doivent être stabilisés à la température ambiante avant de procéder au test. Les échantillons d urine qui contiennent des précipités apparents doivent être filtrés, centrifugés ou stabilisés avant de procéder au test. Echantillons de sérum : Le sérum des patientes ne requiert aucune préparation particulière. Le sérum qui n est pas testé immédiatement doit être stocké dans un réfrigérateur à 2-8 (36-46 F) durant un maximum de 72 heures ou congelé à -20 (-4 F) pendant un maximum de 3 mois. Ne pas congeler et décongeler à plusieurs reprises les échantillons. Ne pas utiliser les échantillons hémolysés de manière grossière. PROÉDURE DE ES Se reporter au paragraphe «Prélèvement des échantillons». S ils ont été congelés ou réfrigérés, les dispositifs de test, les échantillons des patientes et les contrôles doivent être stabilisés à une température ambiante de (68-86 F) avant de procéder au test. Ouvrir les poches juste avant d effectuer le test. 1. Extraire le dispositif de test de sa poche de protection (les dispositifs doivent être stabilisés à température ambiante avant l ouverture de la poche pour éviter la condensation de l humidité sur la membrane). Apposer une étiquette d identification de la patiente et du contrôle sur le dispositif. Placer le dispositif sur une surface plate propre. 2. Extraire l échantillon jusqu au repère sur la pipette (environ 0,2 ml). Préparer la totalité des contenus dans le bac à échantillon.
