1 Tecnologia de Células e Tecidos Ocorrência: 1.º Ano, 2.º Semestre Carga Horária: TP: 45,0; PL: 7,5 Área Científica: Biotecnologia Objetivos de aprendizagem (conhecimentos, aptidões e competências a desenvolver pelos estudantes): Proporcionar aos estudantes uma formação integrada da tecnologia de Células e Tecidos, com base em conceitos fundamentais de Biologia Celular, Histologia, Imunologia e bioprocesso/ bioengenharia, com aplicações na Medicina humana e veterinária. Conteúdos programáticos: 1. Fundamentos de engenharia de células e tecidos: organização de células e tecidos. 2. Métodos de separação e purificação celular e sub-celular. 3. Formação de tecidos: Interações entre célula-matriz, interações células-célula e comunicação celular. 4. Sinalização célula-célula. Transdução de sinal intra-celular. Recetores celulares, mensageiros secundários, cinases/fosfatases. 5. Tecidos epitelial, conjuntivo, nervoso e muscular. 6. Dinâmica e reparação de tecidos. Homeostase. Morfogénese. Diferenciação. 7. Biomateriais na engenharia de tecidos. 8. Microfabricação de tecidos. 9. Conceitos básicos de células estaminais. 10. Reatores para cultura de células animais. Projeto e operação de reatores. Cinética, modelação e monitorização. 11. Processamento de células animais/estaminais. Separação e purificação. Integração de processos de produção e purificação. 12. Aplicações em Medicina Regenerativa: Terapia Celular, Terapia Génica, Engenharia de Tecidos. 1

2 Demonstração da coerência dos conteúdos programáticos com os objetivos de aprendizagem da unidade curricular: Nesta unidade curricular o estudante aprende o modo de organização das células e dos tecidos, tendo em conta o tipo de interações que podem ser estabelecidas entre células e o meio envolvente. O conhecimento de como é feita a diferenciação dos tecidos, bem como os biomateriais usados para a cultura celular e de tecidos e o modo de produção dos mesmos em bio-reator fornece ao estudante as ferramentas usadas na Medicina Regenerativa e Terapia Celular com base na Engenharia dos Tecidos. Os conteúdos programáticos estão por isso de acordo com os objetivos de aprendizagem. Metodologias de ensino (avaliação incluída): A metodologia de ensino assenta numa estratégia dinâmica de aulas teórico/práticas em que os conteúdos teóricos referidos no programa serão lecionados com auxílio de slides de PowerPoint. A exposição teórica é intercalada com componentes de aplicação práticas onde serão apresentados casos estudo, que serão discutidos e analisados pelos estudantes com apoio do docente. A avaliação será feita 70% por 2 frequências (35% cada) e 30% avaliação prática (apresentação e discussão de trabalhos e avaliação contínua do desempenho na aula). A avaliação poderá ainda ser totalmente composta pelo exame final em 1.ª ou 2.ª época (100%). Demonstração da coerência das metodologias de ensino com os objetivos de aprendizagem da unidade curricular: Nesta UC pretende-se que os estudantes expandam o seu conhecimento relativamente às interações entre as células e a formação de tecidos. Para uma melhor consolidação dos conhecimentos intercala-se a exposição dos conteúdos mencionados no programa com a apresentação de case studies, que os estudantes terão que trabalhar e apresentar. A avaliação final desta UC tem em conta a aquisição de conhecimentos teóricos (testes/exame final) e a capacidade de aplicação dos conceitos no tratamento dos case studies referidos. 2

4 Cell and Tissue Technology Calendar: 1 st Year, 2 nd Semester Contact Hours: TP: 45.0; PL: 7.5 Scientific Area: Biotechnology Intended learning outcomes (knowledge, skills and competences to be developed by the students): Provide students with an integrated education of cell and tissue technology, based on fundamental concepts of Cell Biology, Histology, Immunology and bio-process, with applications in human and veterinary medicine. Syllabus: 1. Fundamentals of cell and tissue engineering: organization of cells and tissues. 2. Methods of cell separation and purification, and subcellular. 3. Formation of tissue: cell-matrix interactions, cell-cell interactions, and cellular communication. 4. Cell-cell signalling. Signal intracellular transduction. Cell receptors, second messengers, kinases / phosphatases. 5. Epithelial, connective, muscular and nervous tissue. 6. Tissue dynamics and repair. Homeostasis. Morphogenesis. Differentiation. 7. Biomaterials in tissue engineering. 8. Tissue Microfabrication. 9. Introduction to stem cells. 10. Reactors for animal cell culture. Design and operation of reactors. Kinetics, modelling and monitoring. 11. Animal cells / stem cells processing. Separation and purification. Integration of production processes and purification. 12. Applications in Regenerative Medicine: Cell Therapy, Gene Therapy, Tissue Engineering. 4

5 Evidence of the syllabus coherence with the curricular unit s intended learning outcomes: In this curricular unit, the student learns how cells and tissues are organized, taking into account the type of interactions that can be established between cells and the surrounding environment. The knowledge of how tissue differentiation is made, as well the biomaterials used for cell and tissue culture and the mode of production in a bioreactor provides the student with the tools used in Regenerative Medicine and Cell Therapy based on Tissue Engineering. The contents are therefore according to the learning objectives. Teaching methodologies (including evaluation): The teaching methodologies rely on a dynamic basis of theoretical/practical classes wherein lectures will be given to present the concepts regarding the syllabus, via PowerPoint slides. Practical applications will intercalate, in which case studies will be presented, analyzed, studied and discussed, by the students under the supervision of the teacher. The evaluation will be performed by 70%, consisting in 2 tests (35% each), and 30% practical assessment (presentation and discussion of work and continuous evaluation of performance in class). The evaluation may also be composed entirely of the final exam (100%). Evidence of the teaching methodologies coherence with the curricular unit s intended learning outcomes: In this curricular unit, it is intended that students expand their knowledge regarding the interaction between the cells and tissue formation. For a better consolidation of knowledge, the display of the contents mentioned in the syllabus is merged with the presentation of case studies, which students will have to work and present. The final evaluation of this course takes into account the acquisition of theoretical knowledge (tests / final exam) and the ability to apply the concepts in the treatment of the case studies mentioned. 5
